چکیده

Harmon, Shirley A. (Ohio State University, Columbus), and Jack N. Baldwin. Nature of the determinant controlling penicillinase production in Staphylococcus aureus. J. Bacteriol. 87:593-597. 1964.-From three penicillinase-producing strains of Staphylococcus aureus 38 penicillin-sensitive mutants were isolated. With phages 80 and 53 of the International Typing Series, each of these mutants was a competent recipient of the penicillinase marker from the parent strains. Although numerous reciprocal transductions were performed between penicillin-sensitive mutants, no penicillinase-producing recombinants were observed. Reversion of the penicillinase-negative mutants to capacity to produce penicillinase was never observed. When tested for 50 distinctive physiological characteristics, the mutants generally were similar to the parent strains except for resistance to penicillin. One penicillin-sensitive mutant failed to produce detectable amounts of five exoenzymes. By treating resistant strains with acridine orange, it was possible to isolate sensitive strains at a frequency that was 60 times greater than the spontaneous mutation rate. Thus, the determinant controlling the capacity of strains of S. aureus to produce penicillinase appears to be a cytoplasmic element.

Phenotypic suppression of methicillin resistance in Staphylococcus aureus by mutant noninducible penicillinase plasmids.

Methicillin (intrinsic) resistance of Staphylococcus aureus was suppressed almost completely by regulatory gene (penI(1)) mutations of penicillinase plasmids that made penicillinase production strictly noninducible. Methicillin resistance was restored by secondary regulatory gene mutations that altered the noninducible phenotype or by complementation with a compatible plasmid that did not bear ...

Nature of the genetic determinant controlling exfoliative toxin production in Staphylococcus aureus.

Phage group II Staphylococcus aureus has been identified as the etiological agent of the staphylococcal scaleded skin syndrome. The development of an animal model system permitted fulfillment of Koch's postulates and recognition of exfoliative toxin (ET) as being responsible for some of the clinical manifestations of this syndrome. Initial studies directed toward associating a lysogenic phage w...

Wild-type strain of Staphylococcus aureus containing two genetic linkage groups for penicillinase production.

Staphylococcus aureus strain 55C1, isolated from a patient in 1955, contained two genetic linkage groups for penicillinase formation. One was linked to genes that control resistance to cadmium and mercuric ions; it had properties of a plasmidborne gene. The other was not linked to resistance to these metal ions; it had properties of a chromosomal gene. Penicillinase formation by cells that cont...

Naturally occurring penicillinase plasmids in Staphylococcus aureus.

A series of plasmids harbored by naturally occurring penicillin-resistant strains of Staphylococcus aureus were surveyed with a view toward exploring the variability in plasmid-linked marker patterns. Plasmids were transduced from their natural hosts to either of two plasmid-negative laboratory strains by selection for cadmium resistance, and the transductants were tested for all other markers ...

Nature of the elimination of the penicillinase plasmid from Staphylococcus aureus by surface-active agents.

Growth of Stapylococcus aureus in various ionic surface-active agents resulted in loss of the ability to produce penicillinase, whereas growth in nonionic surface-active agents had no effect on penicillinase production. The curing effect of various alkyl sulfates was found to be dependent upon the chain length. Curing by surface-active agents could be inhibited by magnesium. Reciprocal transduc...
